A viral serotype is an antigenically distinct category within a virus group or species, identified by how antibodies recognize and neutralize it. Viruses of the same serotype show a broadly similar reaction to a defined set of antibodies, whereas another serotype is not neutralized in the same manner. Thus, serotype is an immunological classification rather than merely a classification by genetic sequence.

Basis of serotype distinction

Antibodies recognize specific molecular regions called epitopes, generally on exposed viral capsid or envelope proteins. Differences in these epitopes can alter antibody binding or neutralization and thereby distinguish serotypes.

  • Antibodies generated against one serotype may neutralize another serotype weakly or not at all.
  • Cross-reactive binding does not necessarily imply effective neutralization or protection.
  • Serotype chiefly describes antibody-mediated antigenic differences; it does not by itself indicate disease severity or transmissibility.

How serotypes are identified

Serotyping is based on serological reactions between a virus and known antibodies or antisera, with virus-neutralization assays providing a functional measure of whether the antibodies prevent infection of susceptible cells.

  • Serotyping measures antigenic phenotype, while genotyping compares nucleotide sequences.
  • A single serotype may contain multiple genetic variants or strains.
  • A genetic mutation does not automatically create a new serotype unless it produces a meaningful change in serological recognition or neutralization.

Importance for vaccines

Serotype differences matter because immunity against one serotype may not provide adequate protection against others.

  • Vaccines may need to include antigens from multiple serotypes to generate broad protection.
  • Vaccine evaluation may assess neutralizing-antibody responses separately against the relevant serotypes.
  • Dengue virus has four recognized serotypes, illustrating why balanced protection against several serotypes is important.
  • The presence of antibodies in a binding test alone is not sufficient evidence that those antibodies are protective.
